Title: Platform of mouse and human embryonic stem cell-derived cardiomyocyte test systems for drug development

Objectives: In the ES2HEART a consortium of 3 SME and 3 academic partners will employ already established mouse and human embryonic stem (ES) cell lines to develop innovative methods for the targeted preparation of differentiated cardiac muscle cells and will create an internationally novel test system for pharma/biotech industry and medical costumers. Genetically modified ES cell lines will be prepared to derive cardiomyocytes with well-defined and stabilized characteristics. These cardiomyocytes will then be made available for high-throughput screenings for pharmacological studies including efficacy, safety, and toxicology. Stem cells will be also implanted into the myocardium for testing in vivo cardioprotection and regeneration of myocardial ischemia/reperfusion injury and infarction. The close collaboration of academic and SME partners with internationally recognized expertise in mouse and human ES cell culture, Ca2+-imaging, gene transfer, flow cytometry, confocal microscopy, functional assays and ischemic heart models already established the key technologies for this project. The importance of this approach is demonstrated by the 2007 Medical Nobel Prize awarded for ES cell research.
